Just a tip in advance, do the ambulance missions early on, before you make an enemy of some powerful factions. Same with Police and Taxi.

Ambulance mission not too bad. Do it right after Luigi's Girls before anyone is hostile. Most guides say pick up the furthest ones first since it takes longer but it... never worked for me. I pick up the closest ones first as you can get a minute and a half often from close sucession pick ups giving you leeway like crazy. Square + R1 when vehicle is bobbing and each turn/angled street is your biggest lifesaver. I know the set's not here yet but if you wanna practice I think you'll have an easy time like this

Also, try and go for groups. Pause and think out a route. Great advice @rojopsycho
