Turn turbo on and hold I+II+Select - setting the speed to fast will let you select Hard Human; setting it to slow will let you select Super Mania and God of Game. After selecting difficulty, hold Run and press Select to reset the game.

Dude, don't change the difficulty using turbo otherwise it doesn't work. Instead, alternate between buttons I and II with your fingers and it works.

To change the difficulty level without using turbo button, in intro screen, hold SELECT and press A and B, alternatively, until the difficulty menu shows up. To select SUPER MANIA and GOD OF GAME modes, continue to holding SELECT and pressing A and B, alternatively, in the difficulty menu. Select the difficult with SELECT. Hold RUN and press SELECT to exit.

for anyone trying the difficulty levels, make sure your Mode Switch is set to 2-button controller, NOT 6-button.
